Before treatment
Before you can start MODEYSO, your doctor will confirm if it is right for you
MODEYSO is for adults and children at least 1 year of age.
You may be eligible for MODEYSO if your doctor confirms that you have:
- Diffuse midline glioma
- A specific “H3 K27M” gene mutation
- Body weight of at least 22 pounds (10 kg)
- A tumor that has grown or come back after trying other treatments
Your doctor will perform tests to make sure that MODEYSO is right for you.
If you or the person you care for has heart rhythm problems, including a condition called QT prolongation, or a history of heart problems or heart failure, MODEYSO may not be a suitable treatment. Alert your doctor before starting treatment if you or the person you care for has any of these conditions.
Additional considerations before starting treatment
Pregnancy
If you are a female who may be able to have a child:
- Tell your doctor right away if you become pregnant or think that you may be pregnant during treatment with MODEYSO
- Your doctor should check to see if you are pregnant before you begin treatment with MODEYSO
- Effective birth control should be used while taking MODEYSO and for 1 month after your last dose
If you are a male with a partner who may be able to become pregnant:
- Use effective contraception during treatment with MODEYSO and for 1 month after your last dose
Breastfeeding
If you are breastfeeding or plan to breastfeed:
- As it is not known if MODEYSO passes into breast milk, do not breastfeed during treatment with MODEYSO and for 1 week after your last dose
- Talk to your doctor about the best way to feed your baby during this time
Drug interactions
Tell your doctor about all the medicines you take, including prescription and over-the-counter medicines, vitamins, and herbal supplements.
MODEYSO and other medicines may affect the way each other work and may cause serious side effects. You can ask your doctor or pharmacist for a list of medications that interact with MODEYSO.
Know the medicines you take. Keep a list of your medicines to show your doctor and pharmacist. Do not start or stop taking any new medicines without first talking to your doctor.

How and when to take MODEYSO
Take MODEYSO exactly as your doctor tells you. Do not change your dose or stop taking MODEYSO without talking to your doctor.
How to take MODEYSO
MODEYSO 125-mg capsules should only be taken orally (by mouth). It is important to take the right number of MODEYSO capsules once a week, as instructed by your doctor:
Take MODEYSO capsules when your stomach is empty, either 1 hour before you eat or 3 hours after you’ve eaten
Take all the capsules needed for your dose at the same time, and do this once a week on the same day each week
Swallow each capsule whole with liquid; if you cannot swallow capsules whole, see the Instructions for Use for details on how to prepare and take the contents of the capsules in a suitable drink
If you are giving your child a dose of MODEYSO, the number of capsules in your child’s prescribed dose depends on your child’s weight, and their doctor will tell you how many to give.
MODEYSO should be stored at room temperature (68 °F to 77 °F, or 20 °C to 25 °C)
When to take MODEYSO and what to do if you miss a dose


Additional instructions
- Do not change your dose or stop taking MODEYSO without talking to your doctor
- Your doctor may change your dose, temporarily stop, or completely stop treatment with MODEYSO if you develop certain side effects
- If vomiting happens after taking a dose of MODEYSO, do not take another dose. Instead, wait and take the next dose of once-weekly MODEYSO on the next regularly scheduled day
